Fully Human Anti-TfR1 HCAbs

TfR-1 (transferrin receptor 1), also known as CD71, is a single transmembrane protein that mediates cellular iron uptake by the interaction with iron-bound transferrin (Tf). Expressed on brain endothelial cells, TfR1 can also transport cargos (e.g. drugs with TfR1 targeted) into the brain parenchyma via receptor-mediated transcytosis (RMT). Anti-TfR1 antibodies/VHHs are widely used as blood-brain barrier (BBB) shuttles for brain drug delivery.

    Fully Human Anti-TfR1 HCAbs Highlights

We have developed fully human anti-TfR1 VHHs with

    • Better BBB penetration compared to the JR-141 analog in vivo.
    • Potent brain drug delivery in both bivalent and monovalent format in vivo.
    • Flexible for non-conjugating shuttling, such as fusion proteins or bispecific antibodies, as well as conjugated payload shuttling, including antibody-oligonucleotide conjugates (AOCs) or other small molecule conjugates.
