Gene targeting strategy for B-hCCR5 mice. The exon 2 of mouse Ccr5 gene that encodes the whole molecule (ATG to STOP codon) were replaced by human counterparts in B-hCCR5 mice. The promoter, 5’UTR and 3’UTR region of the mouse gene are retained. The human CCR5 expression is driven by endogenous mouse Ccr5 promoter, while mouse Ccr5 gene transcription and translation will be disrupted.

Strain specific CCR5 expression analysis in homozygous B-hCCR5 mice by flow cytometry. Peritoneal washes were collected from WT and homozygous B-hCCR5 (H/H) mice and analyzed by flow cytometry with species-specific anti-CCR5 antibody. Mouse CCR5 was exclusively detectable in WT mice. Human CCR5 were exclusively detectable in homozygous B-hCCR5 but not WT mice.
