Gene targeting strategy for B-hCDH1 mice.
The exons 4~13 of mouse Cdh1 gene that encode the extracellular domain were replaced by human CDH1 exons 4~13 in B-hCDH1 mice.

Western blot analysis of CDH1 protein expression in homozygous B-hCDH1 mice. Colon tissue lysates were collected from wild-type C57BL/6 (+/+) mice and homozygous B-hCDH1 mice (H/H), and then analyzed by western blot with species-specific anti-CDH1 antibody. 40 μg total proteins were loaded for western blotting analysis. Human CDH1 was detectable in colon of homozygous B-hCDH1 mice.

Strain specific analysis of CDH1 mRNA expression in wild-type C57BL/6 mice and B-hCDH1 mice by RT-PCR. Colon RNA were isolated from wild-type C57BL/6 mice (+/+) and homozygous B-hCDH1 mice (H/H), then cDNA libraries were synthesized by reverse transcription, followed by PCR with mouse or human CDH1 primers. Mouse Cdh1 mRNA was detectable only in wild-type C57BL/6 mice. Human CDH1 mRNA was detectable only in homozygous B-hCDH1 mice but not in wild-type mice.
