Gene targeting strategy for B-hCLEC10A mice.
The exons 3-10 of mouse Mgl2 gene and the exons 3-10 of mouse Mgl1 gene that encodes the full-length protein was replaced by human CLEC10A exons 3-9 in B-hCLEC10A mice.

Strain specific CLEC10A expression analysis in homozygous B-hCLEC10A mice by flow cytometry. BMDC were collected from wild-type C57BL/6 mice (+/+) and homozygous B-hCLEC10A mice (H/H), and analyzed by flow cytometry with species-specific anti-CLEC10A antibody. Mouse Mgl1 and Mgl2 was detectable in wild-type mice. Human CLEC10A was exclusively detectable in homozygous B-hCLEC10A mice but not in wild-type mice.

Strain specific analysis of CLEC10A mRNA expression in wild-type C57BL/6 mice and B-hCLEC10A mice by RT-PCR. Spleen RNA were isolated from wildtype C57BL/6 mice (+/+) and homozygous B-hCLEC10A mice (H/H), then cDNA libraries were synthesized by reverse transcription, followed by PCR with mouse or human CLEC10A primers. Mouse Mgl1 and Mgl2 mRNA were detectable only in wild-type C57BL/6 mice. Human CLEC10A mRNA was detectable only in homozygous B-hCLEC10A mice but not in wild-type mice.
