      Description

      CXCR6 is a receptor for the chemokine CXCL16, which is a 39 kD G-protein coupled chemokine receptor with seven transmembrane-spanning regions.

      Targeting strategy

      Gene targeting strategy for B-hCXCR6 mice. The exons 2 of mouse Cxcr6 gene that encode the whole molecule (ATG to STOP codon were replaced by human counterparts in B-hCXCR6 mice. The promoter, 5’UTR and 3’UTR region of the mouse gene are retained. The human CXCR6 expression is driven by endogenous mouse Cxcr6 promoter, while mouse Cxcr6 gene transcription and translation will be disrupted.

      Protein expression analysis-splenocytes
      Strain specific CXCR6 expression analysis in homozygous B-hCXCR6 mice by flow cytometry. Splenocytes were collected from wild-type C57BL/6 mice (+/+) and homozygous B-hCXCR6 mice (H/H). Protein expression was analyzed with anti-mouse CXCR6 antibody (Biolegend, 151105) and anti-human CXCR6 antibody (Biolegend, 356003) by flow cytometry. mCXCR6 was detectable in T cells, NK cells, and NKT cells of splenocytes in wild-type mice, hCXCR6 was detectable in T cells, NK cells, and NKT cells of splenocytes in homozygous B-hCXCR6 mice.

Protein expression was analyzed with anti-mouse CXCR6 antibody (Biolegend, 151105) and anti-human CXCR6 antibody (Biolegend, 356003) by flow cytometry. mCXCR6 was detectable in T cells and NK cells of liver in wild-type mice, hCXCR6 was detectable in T cells and NK cells of liver in homozygous B-hCXCR6 mice.

      mRNA expression analysis
      Strain specific analysis of CXCR6 mRNA expression in wild-type C57BL/6 mice and B-hCXCR6 mice by RT-PCR. Lung and liver RNA were isolated from wild-type C57BL/6 mice (+/+) and homozygous B-hCXCR6 mice (H/H), then cDNA libraries were synthesized by reverse transcription, followed by PCR with mouse or human CXCR6 primers. Mouse Cxcr6 mRNA were detectable only in wild-type C57BL/6 mice. Human CXCR6 mRNA was detectable only in homozygous B-hCXCR6 mice but not in wild-type mice.
      Protein expression analysis-spleen

Negative Control: APC anti-His Tag Antibody was stained without Mouse or Human CXCL16 Protein (His Tag).
