Gene targeting strategy for B-hEGFL7 mice. The exons 3~10 of mouse Egfl7 gene that encodes the full-length protein was replaced by human EGFL7 exons 4~11 in B-hEGFL7 mice.
Protein expression analysis
Strain specific EGFL7 expression analysis in homozygous B-hEGFL7 mice by ELISA. Serum and lung were isolated from wild-type C57BL/6 mice (+/+) and homozygous B-hEGFL7 mice (H/H), and then analyzed by ELISA with species-specific EGFL7 ELISA kit. Human EGFL7 was exclusively detectable in serum (A) and lung (B) in homozygous B-hEGFL7 mice but not in wild-type mice.
mRNA expression analysis
Strain specific analysis of EGFL7 mRNA expression in wild-type C57BL/6 mice and B-hEGFL7 mice by RT-PCR. Lung RNA were isolated from wild-type C57BL/6 mice (+/+) and homozygous B-hEGFL7 mice (H/H), then cDNA libraries were synthesized by reverse transcription, followed by PCR with mouse or human EGFL7 primers. Mouse Egfl7 mRNA was detectable only in wild-type C57BL/6 mice. Human EGFL7 mRNA was detectable only in homozygous B-hEGFL7 mice but not in wild-type mice.
