Gene targeting strategy for B-hICAM3 mice. The BAC sequences containing 9 kb upstream of human ICAM3 gene, the whole human ICAM3 genomic sequences and 3 kb downstream of the gene were inserted into mouse Rosa26 locus in B-hICAM3 mice.
Strain specific ICAM3 expression analysis in homozygous B-hICAM3 mice by flow cytometry. T cells and B cells from the spleen were collected from wild-type C57BL/6 mice (+/+) and homozygous B-hICAM3 mice (H/H), and analyzed by flow cytometry with species-specific anti-ICAM3 antibodies. Human ICAM3 was exclusively detectable in homozygous B-hICAM3 mice but not in wild-type mice.
