Gene targeting strategy for B-hIL17B mice. The exons 1~3 of mouse Il17b gene that encodes the full-length protein were replaced by human IL17B exons 1~3 in B-hIL17B mice.

Western blot analysis of IL17B protein expression in homozygous B-hIL17B mice. Various tissue lysates were collected from wild-type C57BL/6 mice (+/+) and homozygous B-hIL17B mice (H/H), and then analyzed by western blot with anti-IL17B antibody. 30 μg total proteins were loaded for western blotting analysis. IL17B was detected in liver and trachea in both wild-type and homozygous mice, as the antibody was cross-reactive between human and mouse.

Strain specific analysis of IL17B mRNA expression in wild-type C57BL/6 mice and B-hIL17B mice by RT-PCR. Lung RNA were isolated from wild-type C57BL/6 mice (+/+) and homozygous B-hIL17B mice (H/H), then cDNA libraries were synthesized by reverse transcription, followed by PCR with mouse or human IL17B primers. Mouse Il17b mRNA was detectable only in wild-type C57BL/6 mice. Human IL17B mRNA was detectable only in homozygous B-hIL17B mice but not in wild-type mice.
