      Description
      • Secretory leukocyte protease inhibitor (SLPI) possesses properties that combat inflammation and bacterial infection, while also facilitating the healing process of wounds. In pathological cases, SPLI is also highly expressed in many cancer cell lines, including ovarian, kidney, and colon cancers.
      • The exons 1~3 of mouse Slpi gene that encode the full-length protein were replaced by human SLPI exons 1~4 in B-hSLPI mice.
      • Human SLPI was detectable in lung, liver and kidney of homozygous B-hSLPI mice.
      • Mouse Slpi mRNA was detectable in lung of wild-type C57BL/6 mice. Human SLPI mRNA was detectable in B-hSLPI mice.
      Targeting strategy

      Gene targeting strategy for B-hSLPI mice. The exons 1~3 of mouse Slpi gene that encode the full-length protein were replaced by human SLPI exons 1~4 in B-hSLPI mice.

      mRNA expression analysis
      Strain specific analysis of SLPI gene expression in wild-type mice and B-hSLPI mice by RT-PCR. Mouse Slpi mRNA was detectable in lung of wild-type mice (+/+). Human SLPI mRNA was detectable only in homozygous B-hSLPI mice but not in wild-type mice.
      Protein expression analysis
      Western blot analysis of SLPI protein expression in wild-type mice and B-hSLPI mice. Various tissue lysates were collected from wild-type C57BL/6 (+/+) mice and homozygous B-hSLPI mice(H/H), and then analyzed by western blot with anti-SLPI antibody. 40 μg total proteins were loaded for western blotting analysis.
