    Orthotopic Breast Cancer Model: 4T1 in Balb/c Mice

    Establishment of orthotopic breast cancer model.
    After the inoculation of 4T1 cells in Balb/c mice (F, n=5), the orthotopic tumor can achieve a stable and moderate growth rate, and had no effect on body weight. Flow cytometry showed that there was a significant difference in the number of macrophages and DC cells between orthotopic tumor and subcutaneous tumor. H&E staining showed that tumors metastasized to the lung.

    Orthotopic Breast Cancer Model: MDA-MB-231 in B-NDG Mice

    Establishment of orthotopic breast cancer model.
    MDA-MB-231 cells (1.5x107and 2x107) were mammary pad into B-NDG mice (female, 11-week-old, n=5). Tumor volume and body weight were measured twice a week. (A) Average tumor volume ± SEM. (B) Body weight (Mean± SEM). Volume was expressed in mm3 using the formula: V=0.5 X long diameter X short diameter2. As shown in panel A, MDA-MB-231 cells were able to establish tumors in vivo and can be used for efficacy studies.

    Orthotopic Breast Cancer Model: EO771 in C57BL/6N Mice

    Establishment of orthotopic breast cancer model.
    EO771 cells were subcutaneously (5x104, 1x105) and mammary pad (2x104, 5x104, , 1x105 , 2x105) into C57BL/6 mice (female, 8-week-old, n=5). Tumor volume and body weight were measured twice a week. (A) Average tumor volume ± SEM. (B) Body weight (Mean± SEM). Volume was expressed in mm3 using the formula: V=0.5 X long diameter X short diameter2. As shown in panel A, EO771 cells were able to establish tumors in vivo and can be used for efficacy studies.

    DU4475 cells (2x106, 5x106, 1x107) were subcutaneously and mammary pad into B-NDG B2m KO mice (female, 11-week-old). Tumor volume and body weight were measured twice a week. (A) Average tumor volume ± SEM. (B) Body weight (Mean± SEM). Volume was expressed in mm3 using the formula: V=0.5 X long diameter X short diameter2. As shown in panel A, DU4475 cells were able to establish tumors in vivo and can be used for efficacy studies.
