Gene targeting strategy for B-hCD151 mice.
The exons 3~8 of mouse Cd151 gene that encode the extracellular domains of CD151 protein were replaced by human CD151 exons 3~8 in B-hCD151 mice.

Western blot analysis of CD151 protein expression in homozygous B-hCD151 mice. Various tissue lysates were collected from wild-type C57BL/6 mice (+/+) and homozygous B-hCD151 mice (H/H), and then analyzed by western blot with species-specific anti-CD151 antibody. 30 μg total proteins were loaded for western blotting analysis. hCD151 was exclusively detected in colon, kidney, lung and spleen in homozygous B-hCD151 mice.

Strain specific analysis of CD151 mRNA expression in wild-type C57BL/6 mice and B-hCD151 mice by RT-PCR. Lung RNA were isolated from wild-type C57BL/6 mice (+/+) and homozygous B-hCD151 mice (H/H), then cDNA libraries were synthesized by reverse transcription, followed by PCR with mouse or human CD151 primers. Mouse Cd151 mRNA was detectable only in wild-type C57BL/6 mice. Human CD151 mRNA was detectable only in homozygous B-hCD151 mice but not in wild-type mice.
