      Targeting strategy

      Gene targeting strategy for B-hCD22 mice. The exons 4~11 of mouse Cd22 gene that encode the extracellular domain were replaced by human CD22 exons 2~9 in B-hCD22 mice.

      protein expression analysis

      Strain specific CD22 expression analysis in heterozygous B-hCD22 mice by flow cytometry. Splenocytes were collected from WT and heterozygous B-hCD22 (H/+) mice, and analyzed by flow cytometry with species-specific CD22 antibody. Mouse CD22 was detectable in WT mice and heterozygous B-hCD22 mice. Human CD22 was exclusively detectable in heterozygous B-hCD22 mice but not in WT mice.
