Targeting strategy
Gene targeting strategy for B-hDLK1 mice. The exons 3~5 of mouse Dlk1 gene that encodes the full-length protein was replaced by human DLK1 exons 3~5 in B-hDLK1 mice.
Protein expression analysis
Western blot analysis of DLK1 protein expression in homozygous B-hDLK1 mice. Various tissue lysates were collected from wild-type C57BL/6 mice (+/+) and homozygous B-hDLK1 mice (H/H), and then analyzed by western blot with anti-DLK1 antibody. 1 μg/μL total proteins were loaded for western blotting analysis by JESS. Human DLK1 was detected in embryo, placenta, fetal liver and adrenal gland.
mRNA expression analysis
Strain specific analysis of DLK1 mRNA expression in wild-type C57BL/6 mice and B-hDLK1 mice by RT-PCR. Lung RNA were isolated from wild-type C57BL/6 mice (+/+) and homozygous B-hDLK1 mice (H/H), then cDNA libraries were synthesized by reverse transcription, followed by PCR with mouse or human DLK1 primers. Mouse DLK1 mRNA were detectable in wild-type C57BL/6. Human DLK1 mRNA was detectable in homozygous B-hDLK1 mice.
