Targeting strategy
Gene targeting strategy for B-hDPP3 mice. The exon 2~18 of mouse Dpp3 gene that encodes the full-length protein was replaced by human DPP3 exon 2~18 in B-hDPP3 mice.
Protein expression analysis
Western blot analysis of DPP3 protein expression in homozygous B-hDPP3 mice. Various tissue lysates were collected from wild-type C57BL/6 (+/+) mice and homozygous B-hDPP3 mice (H/H), and then analyzed by western blot with species-specific anti-DPP3 antibody. 40 μg total proteins were loaded for western blotting analysis. hDPP3 was detected in liver,kidney,heart,lung,spleen and thymus from homozygous mice.
mRNA expression analysis
Strain specific analysis of DPP3 mRNA expression in wild-type C57BL/6 mice and B-hDPP3 mice by RT-PCR. Liver RNA was isolated from wild-type C57BL/6 mice (+/+) and homozygous B-hDPP3 mice (H/H), then cDNA libraries were synthesized by reverse transcription, followed by PCR with mouse or human DPP3 primers. Mouse Dpp3 mRNA was detectable only in wild-type mice.Human DPP3 mRNA was detectable only in homozygous B-hDPP3 mice but not in wild-type mice.
