      Gene targeting strategy for B-hSERPINE2 mice. The exons 2~9 of mouse Serpine2 gene that encode the full-length protein were replaced by human SERPINE2 exons 2~9 in B-hSERPINE2 mice.

      mRNA expression analysis

      Strain specific analysis of SERPINE2 gene expression in wild-type mice and B-hSERPINE2 mice by RT-PCR. Mouse SERPINE2 mRNA was detectable in lung of wild-type mice (+/+). Human SERPINE2 mRNA was detectable only in lung of homozygous B-hSERPINE2 but not in wild-type mice.

      Western blot analysis of SERPINE2 protein expression in homozygous B-hSERPINE2 mice. Various tissue lysates were collected from wild-type C57BL/6 mice (+/+) and homozygous B-hSERPINE2 mice (H/H), and then analyzed by western blot with species-specific anti-SERPINE2 antibody. 40 μg total proteins were loaded for western blotting analysis. SERPINE2 was detected in brain, spleen, lung, and ovary.
