Gene targeting strategy for B-hTDGF1 mice. The exons 1-6 of mouse Tdgf1 gene that encode the full-length protein were replaced by human TDGF1 exons 1-6 in B-hTDGF1 mice.

Strain specific analysis of TDGF1 gene expression in wild-type C57BL/6 mice and homozygous B-hTDGF1 mice by RT-PCR. Mouse Tdgf1 mRNA was detectable only in embryos of wild-type C57BL/6 mice (+/+). Human TDGF1 mRNA was detectable only in homozygous B-hTDGF1 mice (H/H), but not in wild-type mice.

Strain specific TDGF1 expression analysis in homozygous B-hTDGF1 mice by western blot. Embryos tissue was collected from wild-type C57BL/6 mice (+/+) and homozygous B-hTDGF1 (H/H) mice, and analyzed by western blot with anti-TDGF1 antibody. TDGF1 was detectable in wild-type mice and homozygous B-hTDGF1 mice due to the cross-reactivity of antibodies.
