      Targeting strategy

      Gene targeting strategy for B-hCD3E/hB7-H4 mice.
      The exons 2-6 of mouse Cd3e gene that encode the extracellular domain were replaced by human CD3E exons 2-7 in B-hCD3E/hB7-H4 mice.
      The exons 3-5 of mouse B7-h4 gene that encode the extracellular region coding sequences were replaced by human B7-H4 exons 3-5 in B-hCD3E/hB7-H4 mice.

      mRNA expression analysis

      Strain specific analysis of B7-H4 gene expression in wild type (WT) mice and B-hCD3E/hB7-H4 mice by RT-PCR. Mouse B7-h4 mRNA was detectable only in lung of WT mice (+/+). Human B7-H4 mRNA was detectable only in homozygous B-hCD3E/hB7-H4 mice (H/H) but not in WT mice (+/+).

      Protein expression analysis

      Strain specific CD3E/hB7-H4 expression analysis in homozygous B-hCD3E/hB7-H4 mice by western blot. Lung, uterus, heart, kidney and breast were collected from wild type (WT) mice (+/+) and homozygous B-hCD3E/hB7-H4 mice (H/H), and analyzed by western blot with anti-B7-H4 antibody. B7-H4 was detectable in WT mice (+/+), homozygous B-hCD3E/hB7-H4 mice (H/H) and human B7-H4 positive cell line SK-BR-3 due to the cross-reactivity of antibodies.

      Strain specific CD3E expression analysis in homozygous B-hCD3E/hB7-H4 mice by flow cytometry. Splenocytes were collected from wild type (WT) mice (+/+) and homozygous B-hCD3E/hB7-H4 mice (H/H), and analyzed by flow cytometry with species-specific anti-CD3E antibody. Mouse CD3E was detectable in WT mice (+/+). Human CD3E was exclusively detectable in homozygous B-hCD3E/hB7-H4 mice (H/H) but not in WT mice (+/+).
