Erythropoietin R is a transmembrane protein expressed on the surface of megakaryocytes, erythroid progenitors, and endothelial cells. It binds Erythropoietin and transmits signals that stimulate the proliferation and maturation of bone marrow erythroid precursors into red cells.
The exons 1~6 of mouse Epor gene that encode the extracellular domain and partial transmembrane region were replaced by human EPOR exons 1~6 in B-hEPOR mice.
The expression of EPOR gene was the highest in the erythrocyte colony-forming unit cells and proerythroblasts, and the expression level gradually decreased with cell differentiation, but was not expressed in the reticulocyte and mature erythrocyte stage.
Human EPOR was detectable on proerythroblasts, basophilic erythroblasts and polychromatic erythroblasts from bone marrow of B-hEPOR mice but not in wild-type mice.
Human EPOR mRNA was detectable in the liver of B-hEPOR mice.

Strain specific EPOR expression analysis in B-hEPOR mice by flow cytometry. Bone marrow cells were collected from wild-type C57BL/6 mice (+/+) and B-hEPOR mice (H/H) , and analyzed by flow cytometry with species-specific anti-human EPOR antibody. Human EPOR was detectable on proerythroblasts, basophilic erythroblasts and polychromatic erythroblasts in B-hEPOR mice only.
